The present study aimed to investigate the potential nephroprotective effect of Sa-khan (<em>Piper</em>, Piperaceae) against high glucose-induced cytotoxicity in a human proximal tubular epithelial cell line. The cytotoxicity induced by high glucose in HK-2 cells, a human proximal tubular epithelial cell line, was studied using the 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide (MTT) assay. The wound scratch assay was performed to evaluate the migration capability of HK-2 cells. Treating cells with high glucose significantly reduced the&nbsp;viability of HK-2 cells (<em>p </em>&lt; 0.001). Sa-khan crude extract (SCE) at concentration of 0.031&nbsp;mg/mL was found to increase the viability of high glucose-induced HK-2 cells (<em>p </em>= 0.013). In addition, high glucose significantly increased the migration capability of HK-2 cells (<em>p </em>&lt; 0.001). Furthermore, SCE at concentrations of 0.016 and 0.031 mg/mL abolished the increased migration capability of high glucose-induced cells (<em>p </em>&lt; 0.001). This study shows that Sa-khan crude extract displays a potential protective effect against high glucose-induced proximal tubular cell injury.</p> ER -
