Estrogenic and Antiestrogenic Effects of Methanol Extract of Persea americana Pulp on the Uterus of Immature Female Albino Wistar Rat

Abstract
Estrogen is important for maturation and maintenance of female reproductive health. The aim is to investigate the estrogenic and antiestrogenic effect of Persea americana pulp on immature female Wistar rats using ovariectomized animals. Exactly 45 immature female rats randomly divided into 9 groups of five animals each were used. Ovariectomy was done for 40 rats. Group 1 served as the control, group 2 received distilled water as the sham operated group, group 3 received 0.1mg/kg of 17ß estradiol, groups 4, 5, and 6 received 380mg/kg, 720mg/kg, and 1100mg/kg of Persea americana respectively. These doses of the extract were co-administered with 17ß Estradiol in groups 7, 8, and 9 respectively. Doses were determined from the acute toxicity studies using the Up and Down method. Blood was collected for hormonal analysis and the organs were harvested for organ morphometry and histological analysis after 8 days of administration. The uterine weight of Persea americana + estradiol treated groups significantly increased whereas the body weight decreased compared to group 3. Estrogen and FSH significantly increased in groups 7, 8, and 9 compared to estradiol treated group. Estrogen significantly increased in Persea americana treated groups when compared to group 2. There was evidence of the cuboidal epithelium being stratified, hypertrophy of the endometrial glands, and thickened endometrium in both Persea americana treated groups and Persea americana + estradiol treated groups compared to groups 2 and 3 respectively. It could be concluded that methanol extract of Persea americana is a potential source for estrogen
